A Stimulator Protein of Rubber Biosynthesis from Hevea brasiliensis Latex
نویسندگان
چکیده
A rubber biosynthesis stimulator protein (RBSP)from the C-serum of Hevea brasiliensis latex was purified and characterised. This proteinaceous factor stimulates the incorporation of isopentenyl diphosphate into rubber and thereby enhances the rate of rubber biosynthesis. The heat stable RBSP is shown to have a molecular weight of 13 068 Da by mass spectroscopy. Amino acid sequence of peptide fragments obtained from trypsin digestion revealed regions of sequence similar to eukaryotic Initiation Factor 5 A (eIF-5A), present in alfalfa (Medicago saliva), tobacco (Nicotiana tabacum and Nicotiana plumbaginifolia), Arabidopsis thaliana and potato (Solarium tuberosumj. Three Hevea eIF-5A clones were obtained from screening a latex cDNA library with a partial eIF-5A cDNA from tobacco. Small variations in their DNA sequences indicated that they represented three different isoforms.

Molecular cloning, expression and characterization of cDNA encoding cis-prenyltransferases from Hevea brasiliensis. A key factor participating in natural rubber biosynthesis.
Natural rubber from Hevea brasiliensis is a high molecular mass polymer of isoprene units with cis-configuration. The enzyme responsible for the cis-1,4-polymerization of isoprene units has been idengified as a particle-bound rubber transferase, but no gene encoding this enzyme has been cloned from rubber-producing plants. By using sequence information from the conserved regions of cis-prenyl c...
متن کاملInsights into rubber biosynthesis from transcriptome analysis of Hevea brasiliensis latex.
Hevea brasiliensis is the most widely cultivated species for commercial production of natural rubber (cis-polyisoprene). In this study, 10,040 expressed sequence tags (ESTs) were generated from the latex of the rubber tree, which represents the cytoplasmic content of a single cell type, in order to analyse the latex transcription profile with emphasis on rubber biosynthesis-related genes. A tot...

Rubber (cis-1,4-polyisoprene), an important raw material for many industrial uses, is synthesized in the latex of Hevea brasiliensis (Willd. ex A. Juss.) Müll. Arg. We postulated that the genes uniquely or preferentially expressed in the latex may be important for rubber biosynthesis. We constructed cDNA libraries from the latex of H. brasiliensis to investigate the genes expressed in the latex...

In plants MADS-box transcription factors (TFs) play important roles in growth and development. However, no plant MADS-box gene has been identified to have a function related to secondary metabolites regulation. Here, a MADS-box TF gene, designated as HbMADS4, was isolated from Hevea brasiliensis by the yeast one-hybrid experiment to screen the latex cDNA library using the promoter of the gene e...
متن کاملThe rubber tree genome shows expansion of gene family associated with rubber biosynthesis
Hevea brasiliensis Muell. Arg, a member of the family Euphorbiaceae, is the sole natural resource exploited for commercial production of high-quality natural rubber. The properties of natural rubber latex are almost irreplaceable by synthetic counterparts for many industrial applications. A paucity of knowledge on the molecular mechanisms of rubber biosynthesis in high yield traits still persis...
